본문 1. 실험요약 (1) 실험 방법 ① 미리 배양한 대장균 (E.coli) 세포 1.9㎖을 2㎖ tube에 넣고 13,000rpm으로 10분간 원심분리 한 후 상등액은 버린다. ② 남은 용액은 Micro pipette으로 제거하고 Cell 덩어리를 얻는다. 이때 배지 성분이 남아있지 않도록 pipette으로 배지를 완전히 제거한다. ③ Cell이 들어있는 2㎖ tube에 S1 solution 250㎕을 넣고 pipetting한다. ④ S2 solution 250㎕을 넣고 조심스럽게 3~4번 inverting을하고 2분간 방치한다. ⑤ S3 solution 350㎕ 조심스럽게 inverting 한다. 참고문헌 (1) http://en.wikipedia.org/wiki/DNA_ligase (2)http://blog.naver.com/withcoffee67?Redirect=Log&logNo=70160635571 하고 싶은 말 참고자료로 활용하셔서 좋은 결과 있기를 바랍니다.
